Doom monsters have this sexy blend of demonic Lovecraftian/mythological elements, injected with cybernetic hardware and guns. There's a lot of awesome monster ideas to take away from things like Dungeons and Dragons, H. P. Lovecraft, ancient mythology, and other sci-fi/fantasy lore. Many of these things are often seen as these paralyzing terrifying unstoppable monstrosities that should be feared, but usually because the setting they are found in is some sort of horror or dark ages context where the protagonist is not a badass spacemarine with a ton of firepower. Ghosts, Dragons, Werewolves, Giants, Mummies, Bats, Seamonsters, Demons, Witches, Aliens, Mutants, and more! What kind of horrible abominations, whether it be in other games, art, books, movies, or whatever would you love to destroy with a supershotgun, blast with a rocket launcher, or obliterate with a BFG? HydrasByakheesOrcsOwlbearsCyclops(this is meant purely for imagination masturbation, not necessarily a practical discussion about whether these monsters would work, look good as sprites, or would actually be fun to fight in any capacity in the game.)

Hydra would be a good fight; say, three different heads that spit three different attacks, three separate hit boxes so you get a choice between which attack to nerf first.
Also, I always wanted to see some plant-based enemies, something akin to the muncher plants from super mario. Mostly stationary turrets. I made one once, sprites and dehacked, many moons ago, but it turned out that stationary enemies were one thing that vanilla doom really didn't do well at all.
Maybe something suicide bombery, charges you and then detonates at close range / when you kill it. You could mix them in with other pack enemies to encourage target prioritization / chain reaction shenanigans.

Best idea for an enemy I had, back in the days when I used to daydream TC's, was a Seedship AI.
Basically, the mission was set on board an ark that had fled Earth with a small population in stasis. The ship's computer had access to DNA synthesizers; it went mad, decided it was God, (in the beginning there was darkness and I moved upon the darkness yadda yadda yadda) and started making all sorts of crazy fucking shit from recombined DNA strands.
Because God creates life, yo.
Anyway, the lesser enemies would have just been your standard crazy mutant shit, but the Big Bad would be this HAL super computer that spoke to you as you moved through the ship, spouting philosophy, musing about the survival of the fittest, being a mad bastard in general. The finale would have been moving from room to room shooting memory banks (50% computer, 50% the zebetite things in Metroid before Mother Brain) and with each memory bank destroyed the computer would get more and more retarded until it regressed to infancy, goo-ing and gah-ing and making the player feel REALLY FUCKING TERRIBLE for killing it.
Then the ship explodes and everyone dies anyway. "Let there be light."
It was a total rip off of Kubrick, but in Doom. And then Portal happened, and that was another great idea consigned to the dustbin of history.
But yeah. The best enemy ever would be HAL with a God Complex.

You could achieve this is in Boom format though if you can compromise. First thing that comes to mind is Valiant’s Map30 with the turret enemies. You could probably check out the dehacked for Valiant to see how it functions.

I've always thought Beastmen from Warhammer would be a great, fearsome, opponent to fight aesthetically.. especially if we used the duel ax armored variation of a bestigor.. only challenge would be finding a combat role for it that doesn't render something else redundant.
